Role of CRH in colitis and colitis-associated cancer: a combinative result of central and peripheral effects?

Corticotropin-releasing factor family peptides (CRF peptides) comprise corticotropin releasing hormone (CRH), urocortin (UCN1), UCN2 and UCN3. CRH is first isolated in the brain and later with UCNs found in many peripheral cells/tissues including the colon. CRH and UCNs function via the two types of receptors, CRF1 and CRF2, with CRH mainly acting on CRF1, UCN1 on both CRF1 &CRF2 and UCN2-3 on CRF2. Compiling evidence shows that CRH participates in inflammation and cancers via both indirect central effects related to stress response and direct peripheral influence. CRH, as a stress-response mediator, plays a significant central role in promoting the development of colitis involving colon motility, immunity and gut flora, while a few anti-colitis results of central CRH are also reported. Moreover, CRH is found to directly influence the motility and immune/inflammatory cells in the colon. Likewise, CRH is believed to be greatly related to tumorigenesis of many kinds of cancers including colon cancer via the central action during chronic stress while the peripheral effects on colitis-associated-colon cancer (CAC) are also proved. We and others observe that CRH/CRF1 plays a significant peripheral role in the development of colitis and CAC in that CRF1 deficiency dramatically suppresses the colon inflammation and CAC. However, up to date, there still exist not many relevant experimental data on this topic, and there seems to be no absolute clearcut between the central and direct peripheral effects of CRH in colitis and colon cancer. Taken together, CRH, as a critical factor in stress and immunity, may participate in colitis and CAC as a centrally active molecule; meanwhile, CRH has direct peripheral effects regulating the development of colitis and CAC, both of which will be summarized in this review.

Role of CRF 2 Receptors in Colonic Ion Secretion in Response to Stress‐related Peptides

Background and Aims Stress increases intestinal secretion and exacerbates symptoms of irritable bowel syndrome (IBS). Peripherally-derived corticotropin-releasing factor (CRF) is known to mediate stress-induced intestinal secretion, presumably by activation of CRF1 receptors in the gut. The present study aimed to ascertain the role of CRF2 activation in intestinal secretion by three other members of CRF peptide family, urocortin (UCN) 1-3, in wild-type (WT) and CRF2 knockout (Crhr2-/-) mice. Methods Mucosal/submucosal preparations from proximal colon of WT and Crhr2-/- mice of adult male mice were mounted in Ussing chambers for measurement of short-circuit current (Isc) as an indicator of ion secretion. Results The colonic mucosa/submucosa preparations were exposed to different concentrations of UCN1, 2, or 3 (30 nM, 100 nM, 300 nM, and 1 mM) on the basolateral side. All three UCNs evoked increases in colonic ISC (ΔISC) in a dose-dependent manner. The EC50s were 392.64nM for UCN1, 103.28nM for UCN2, and 231.21nM for UCN3. Application of CRF or stressin1 (a selective CRF1 receptor agonist) (30nM – 1mM) also caused increases in ISC with an EC50 of 597.04nM or 588.84nM, respectively. The potency order was therefore UCN2 > UCN3 > UCN1 > stressin1 » CRF. At each concentration tested, UCN3 produced significantly greater ΔISC responses than that caused by stressin1, CRF, UCN1, or UCN2. Involvement of the CRF receptor subtypes in UCN1-3 evoked ΔISC responses was investigated with the selective CRF1 receptor antagonist NBI27914 and the selective CRF2 receptor antagonist antisauvagine-30. Both NBI27914 and antisauvagine-30 significantly suppressed UCN1 (300nM)-induced ΔISC responses (p < 0.05). The ΔISC responses evoked by UCN2 (100nM) and UCN3 (100nM) were significantly suppressed by antisauvagine-30 (p < 0.05 and p < 0.0001, respectively), but were not influenced by NBI27914 (p > 0.05). In the Crhr2-/- mice, UCN2 (1mM) failed to evoke any ΔISC compared with WT controls (p < 0.01). UCN3 (1mM)-evoked ΔISC responses were significantly decreased by 57.21% in the Crhr2-/- mice compared with WT mice (p < 0.05). In contrast, stressin1 (1mM)-evoked ΔISC response did not differ between WT and Crhr2-/- mice (p > 0.05). Conclusions UCN1-3 all caused an increase of colonic ion secretion, which mimicked the effects of stress and CRF. UCN1-induced increase of colonic ion secretion was mediated by both CRF1 and CRF2, whereas UCN2 and UCN3-induced increase of colonic ion secretion was mediated only by CRF2. Since UCN2 and UCN3 have greater potency and efficacy on colonic ion secretion compared with UCN1, CRF, and stressin1, it is logical to conclude that CRF2 receptors played a predominant role in the pro-secretory effects of UCNs.

Effects of stress-related peptides on chloride secretion in the mouse proximal colon.

Stress increases intestinal secretion and exacerbates symptoms of irritable bowel syndrome (IBS). The present study aimed to ascertain the role of CRF2 activation in intestinal secretion by three other members of CRF peptide family, urocortin (UCN) 1-3, in wild type (WT) and CRF2 knockout (Crhr2-/- ) mice. Mucosal/submucosal preparations from proximal colon of WT and Crhr2-/- mice of both sexes were mounted in Ussing chambers for measurement of short-circuit current (Isc ) as an indicator of ion secretion. Male mice demonstrated a significantly higher baseline Isc than female in both WT and Crhr2-/- genotypes. CRF and UCN1-3 (1μM) caused greater increases in colonic Isc (ΔIsc ) in male than female. Colonic Isc response to the selective CRF1 agonist, stressin1, was similar in both sexes. In male mice, the selective CRF2 agonists (UCN2 and UCN3) caused significantly greater ΔIsc than CRF and stressin1. UCN2- and UCN3-evoked ΔISC was significantly reduced in preparations pretreated with the selective CRF2 antagonist antisauvagine-30 and in Crhr2-/- mice. The prosecretory effects of urocortins were due to increases in Cl- secretion and involved enteric neurons and mast cells. The findings revealed sex differences in baseline colonic secretion and responses to stress-related peptides. CRF2 receptors play a more prominent role in colonic secretion in male mice. The greater baseline secretion and responses to UCNs may contribute to the higher prevalence of diarrhea-predominant IBS in males.

Corticotropin-Releasing Factor: An Ancient Peptide Family Related to the Secretin Peptide Superfamily.

Corticotropin-releasing factor (CRF) is the hypothalamic releasing peptide that regulates the hypothalamic-pituitary-adrenal/inter-renal (HPA/I) axis in vertebrates. Over the last 25 years, there has been considerable discussion on its paralogs genes, urotensin-I/urocortin-1, and urocortins-2 and-3 and their subsequent role in the vertebrate stress response. Phylogenetically, the CRF family of peptides also belong to the diverse assemblage of Secretin- and Calcitonin-based peptides as evidenced by comparative-based studies of both their ligand and G-protein-coupled receptor (GPCR) structures. Despite this, the common origin of this large assemblage of peptides has not been ascertained. An unusual peptide, teneurin-C-terminal associated peptide (TCAP), reported in 2004, comprises the distal extracellular tip of the teneurin transmembrane proteins. Further studies indicated that this teneurin region binds to the latrophilin family of GPCRs. Initially thought to be a member of the Secretin GPCR family, evidence indicates that the latrophilins are a member of the Adhesion family of GPCRs and are related to the common ancestor of both Adhesion and Secretin GPCR families. In this study, we posit that TCAP may be a distantly related ancestor of the CRF-Calcitonin-Secretin peptide family and evolved near the base of metazoan phylogeny.

Information Processing in Affective Disorders: Did an Ancient Peptide Regulating Intercellular Metabolism Become Co-Opted for Noxious Stress Sensing?

Affective disorders arise in stressful situations from aberrant sensory information integration that affects energetic nutrient (i.e., glucose) utilization to the cognitive centers of the brain. Because energy flow is mediated by molecular signals and receptors that evolved before the first complex brains, the phylogenetically oldest signaling systems are essential in the etiology of affective disorders. The corticotropin-releasing factor (CRF) peptide subfamily is a phylogenetically old metazoan peptide family and is pivotal for regulating organismal energy response associated with stress. Highly conserved, both the CRF peptide family and its receptors possess a structural relationship to the teneurins, and their receptors, latrophilins, respectively. The CRF homologous region of teneurin is defined as the "teneurin C-terminal associated peptide" (TCAP) and antagonizes CRF action, regulates mitochondrial energy production, and is anxiolytic in vivo. Here, it is postulated that TCAP represents an ancient peptide that mediates intercellular information transfer of stressful and noxious events by regulating energy utilization among neurons.

Glucocorticoid withdrawal affects stress-induced changes of urocortin 2 gene expression in rat adrenal medulla and brain.

Corticotropin-releasing factor is well known activator of the hypothalamic-pituitary-adrenocortical axis, that represents crucial system participating on stress response of the organism. Urocortins are members of the corticotropin-releasing factor family of peptides with proposed effects on neuroendocrine and behavioral stress response mechanisms. Urocortin 2, one of three known urocortins, is present in central and peripheral stress response system and its expression can be augmented by glucocorticoids. In the present study we have examined how glucocorticoid withdrawal affects urocortin 2 gene expression after acute immobilization in the adrenal medulla and selected brain areas in rats. We used pharmacological adrenalectomy to block synthesis of corticosterone. Our results show that the immobilization-induced rise in urocortin 2 mRNA levels in rat adrenal medulla was not inhibited by glucocorticoid withdrawal. On the other hand, observed changes in the brain indicate that the effect of stress and pharmacological adrenalectomy on urocortin 2 gene expression is site-specific. While in the paraventricular nucleus and locus coeruleus the immobilization induced rise of urocortin 2 was not inhibited by pharmacological adrenalectomy in the arcuate nucleus and central amygdala it was. Moreover, we have seen a significant depletion of urocortin 2 plasma levels after immobilization. The immobilization induced rise of urocortin 2 gene expression in rat adrenal medulla and brain areas regulating stress response pathways and preservation of its induction after adrenalectomy suggests a role of urocortin 2 in the neuroendocrine stress response of an organism. Corticotropin-Releasing Factor Receptors Modulate Oxytocin Release in the Dorsolateral Bed Nucleus of the Stria Terminalis (BNST) in Male Rats

The neuropeptide oxytocin (OT) plays an important role in the regulation of social and anxiety-like behavior. Our previous studies have shown that OT neurons send projections from the hypothalamus to the dorsolateral bed nucleus of the stria terminalis (BNSTdl), a forebrain region critically involved in the modulation of anxiety-like behavior. Importantly, these OT terminals in the BNSTdl express presynaptic corticotropin releasing factor (CRF) receptor type 2 (CRFR2). This suggests that CRFR2 might be involved in the modulation of OT release. To test this hypothesis, we measured OT content in microdialysates collected from the BNSTdl of freely-moving male Sprague-Dawley rats following the administration of a selective CRFR2 agonist (Urocortin 3) or antagonist (Astressin 2B, As2B). To determine if type 1 CRF receptors (CRFR1) are also involved, we used selective CRFR1 antagonist (NBI35965) as well as CRF, a putative ligand of both CRFR1 and CRFR2. All compounds were delivered directly into the BNSTdl via reverse dialysis. OT content in the microdialysates was measured with highly sensitive and selective radioimmunoassay. Blocking CRFR2 with As2B caused an increase in OT content in BNSTdl microdialysates, whereas CRFR2 activation by Urocortin 3 did not have an effect. The As2B-induced increase in OT release was blocked by application of the CRFR1 antagonist demonstrating that the effect was dependent on CRFR1 transmission. Interestingly, CRF alone caused a delayed increase in OT content in BNSTdl microdialysates, which was dependent on CRF2 but not CRF1 receptors. Our results suggest that members of the CRF peptide family modulate OT release in the BNSTdl via a fine-tuned mechanism that involves both CRFR1 and CRFR2. Further exploration of mechanisms by which endogenous OT system is modulated by CRF peptide family is needed to better understand the role of these neuropeptides in the regulation of anxiety and the stress response.

Energy metabolism and behavior in the corticotropin-releasing factor family of peptides.

This year, 2015, will mark the sixtieth anniversary of the seminal work by Guillemin and Rosenberg (1955) and Schally and Saffran (1955) which, along with the earlier work from Geoffrey Harris' lab, initiated the search for an adrenocorticotropin releasing factor that culminated with the discovery of corticotropin-releasing factor (CRF) in 1981 by Wylie Vale's laboratory (Vale et al., 1981). Since the 1980s, the CRF story has had many twists and turns from the finding that CRF and its receptors are located in many extra-hypothalamic brain areas and extending to the discovery that two genome duplications expanded the CRF peptide family hundreds of millions of years ago. Because of the early metazoan ancestry of CRF, this peptide has become ensconced in a number physiological processes. We now know that stress in vertebrates is comprised of a complex set of physiological actions that regulate organismal metabolism to promote protection of the individual and progeny from life-threatening situations. This stress response likely evolved before the earliest multicellular organisms yet has been retained throughout metazoan evolution. Vertebrates, in particular, have developed some of the most complex stress-regulating mechanisms. Integral to this stress response is CRF, and its paralogue, urocortin (urotensin-I/sauvagine) and a parallel paralogous lineage consisting of urocortin 2 and 3. Six review papers in this volume summarize some of the latest research on the role of CRF and urocortin (UCN) peptides in modulating behavior during stress. In addition, two new research studies present data supporting an ancient role for CRF/UCN receptors in social behavior and feeding, while two additional papers explore the role of extrahypothalamic CRF neurons in behavioral and endocrine responses. A consistent theme running through this set of papers is that many of the effects of CRF/UCN peptides on physiology and behavior are evolutionarily conserved. In their contribution, Chen et al. (2013) examine the evolutionary origins of the tenuerin C-terminal associated peptides (TCAP), peptides which may have predated the origin of the CRF peptide family. Chen et al. (2013) review data on the neuromodulatory actions of TCAP-1 and both its direct and indirect effects as an indirect modulator of CRF intracellular signaling. Feeding and intake of nutrients is essential for the success of any species. A stress-response, as regulated by the CRF family of peptides, generally has suppressive actions on appetite and digestion in order to shunt energy from a parasympathetic need to arousal and sympathetic requirements. From an evolutionary point of view this may be an adaptive response when an animal is threatened by a threat in its environment, however there are obvious energetic costs if anorexia is prolonged. As the papers by Matsuda (2013) and Ortega et al. (2013) indicate, CRF inhibition of appetite is a phylogenetically ancient response based on data in fishes. Matsuda (2013) summarizes the data on CRF and appetite in fish and points out the complexities of dissociating specific effects on appetite from the broader impact of CRF peptides on anxiety and locomotor behavior. Ortega et al. (2013) present new data in rainbow trout (Oncorhyncus mykiss) suggesting that the anorexic effects of serotonin may be mediated by CRF- and/or urotensin I receptors. Stengel and Tache (2014) provide an extensive and critical look at CRF/UCN peptide signaling via CRF R1/2 receptors in feeding behavior in mammals. These authors not only summarize the latest studies on the central actions of CRF peptides related to food intake in the context of stress adaptation but also look at the interesting possibility that peripheral receptors also may be involved. Stengel and Tache (2014) also summarize studies on CRF receptor single nucleotide polymorphisms (snps) that are associated with obesity. Emerging evidence from comparative and preclinical studies indicates that CRF may play an adaptive role in modulating social behavior. In vertebrates, social stress is one of the greatest forms of stressors. Prolonged and intense social stress can affect nutrient acquisition and reproduction thus compromising both the health of the individual and the population. In their paper, Solomon-Lane et al. (2013) review the literature linking the hypothalamus-pituitary-interrenal axis to socially-mediated sex change in fishes and provide new data on the role of socially mediated sex change in the blue banded goby (Lythyrpnus dalli). Backstrom and Winberg (2013) take a broad comparative approach to examine the role of the HPI/HPA axis on social stress while Hostetler and Ryabinin (2013) provide an extensive review of the role (s) that central CRF pathways play in social and particularly, reproductive and courtship behavior. Interestingly, a number of authors have commented on the close relationship between the CRF system and the regulation of 5-HT, a neurotransmitter whose evolutionary origins predate multicellular organisms. Elaboration of the central nervous system is a hallmark of vertebrate morphology and success. It is not surprising, therefore, that the CRF peptide family is associated with numerous neurological circuits regulating most behaviors. The role of CRF and its regulation of pituitary ACTH have been well covered in the past. However, several of the papers in this volume touch on the involvement of CRF/UCN peptides targeting receptors in extra hypothalamic brain areas. These studies collectively highlight the diverse phenotypes of cells producing CRF within the CNS. Fox and Lowry (2013) review the interaction between CRF- and 5-HT-producing neurons in the dorsal raphe nucleus, the major site for 5-HT production within the brain. Dabrowska et al. (2013) provide evidence that CRF neurons in the BNST and PVN are phenotypically different based upon their physiological response to local neurotransmitter release and whether they also synthesize GABA or glutamate and may have either inhibitory or excitatory actions, respectively. Callahan et al. (2013) use RNAi techniques to locally knockdown CRF expression in the central nucleus of the amygdala. Interestingly, disruption of CRF synthesis in this brain area interferes with stress-induced HPA activity and the expression of anxiety-like behaviors during stress. In summary, papers within this compendium illustrate the long evolutionary history of the CRF family of peptides and their importance to organismal physiology and behavior. Despite the achievements over the last 60 years, there remains much to learn about the role of the CRF family of peptides in the physiology and pathology of vertebrates.

Quantitative comparison of mRNA level of corticotropin- releasing factor peptide family in normal kidney and corresponding clear cell renal cell carcinoma specimens.

466 Background: Urocortin (Ucn), a 40 amino acid peptide, belongs to the corticotropin releasing factor (CRF) family and exerts its actions mainly in the periphery through activation of two CRF receptors (CRFRs), CRFR1 and CRFR2 and a binding protein (CRFBP/CRHBP). Both receptors are G-protein coupled and binding of Ucn leads to activation of cAMP-dependent protein kinases via elevation of cAMP levels. Ucn and Urocortin III (UcnIII) are involved in conditions such as regulation of local inflammation, angiogenesis, and inhibition of proliferation. Suppression of neovascularization and inhibition of tumor cell cycling by Urocortins is modulated through CRFRs. Activation of CRFRs by e.g. Ucn inhibits angiogenesis via reduction of vascular endothelial growth factor (VEGF) and suppresses the cell proliferation. Here we characterized the whole CRF family on mRNA levels quantitatively comparing the normal and clear cell carcinoma of the kidney (ccRCC). Methods: In this study we measured the mRNA level of Ucn, UcnIII, CRFR1, CRFR2 and CRHBP in 78 RCC samples and paired histologically normal appearing tissues using quantitative PCR. Statistical analyses were carried out using univariate logistic regression analysis. Results: We found tumour specific down regulation of mRNA expression of UcnIII, CRFR1, CRFR2 and CRHBP in samples of RCCs with clear cell histology compared to paired normal tissues (P&lt;0.01 for all targets). Only Ucn did not show any expression difference between two groups (p=0.17). Conclusions: For the first time we showed the expression profile of the whole CRF peptide family in ccRCC compared to normal kidney on mRNA level. Our data underlines the malfunction of this actually strong protective and anticancer system in renal malignancy and shows the involvement of CRF system in carcinogenesis in kidney. More studies are necessary to find out the detailed roles of the CRF system in renal carcinoma.

Cell type-specific modifications of corticotropin-releasing factor (CRF) and its type 1 receptor (CRF1) on startle behavior and sensorimotor gating

The corticotropin-releasing factor (CRF) family of peptides and receptors coordinates the mammalian endocrine, autonomic, and behavioral responses to stress. Excessive CRF production has been implicated in the etiology of stress-sensitive psychiatric disorders such as posttraumatic stress disorder (PTSD), which is associated with alterations in startle plasticity. The CRF family of peptides and receptors mediate acute startle response changes during stress, and chronic CRF activation can induce startle abnormalities. To determine what neural circuits modulate startle in response to chronic CRF activation, transgenic mice overexpressing CRF throughout the central nervous system (CNS; CRF-COECNS) or restricted to inhibitory GABAergic neurons (CRF-COEGABA) were compared across multiple domains of startle plasticity. CRF overexpression throughout the CNS increased startle magnitude and reduced ability to inhibit startle (decreased habituation and decreased prepulse inhibition (PPI)), similar to previous reports of exogenous effects of CRF. Conversely, CRF overexpression confined to inhibitory neurons decreased startle magnitude but had no effect on inhibitory measures. Acute CRF receptor 1 (CRF1) antagonist treatment attenuated only the effects on startle induced by CNS-specific CRF overexpression. Specific deletion of CRF1 receptors from forebrain principal neurons failed to alter the effects of exogenous CRF or stress on startle, suggesting that these CRF1 expressing neurons are not required for CRF-induced changes in startle behaviors. These data indicate that the effects of CRF activation on startle behavior utilize an extensive neural circuit that includes both forebrain and non-forebrain regions. Furthermore, these findings suggest that the neural source of increased CRF release determines the startle phenotype elicited. It is conceivable that this may explain why disorders characterized by increased CRF in cerebrospinal fluid (e.g. PTSD and major depressive disorder) have distinct symptom profiles in terms of startle reactivity.

CRF family peptides are differently altered by acute restraint stress and chronic unpredictable stress

Corticotropin-releasing factor (CRF) acts to promote stress-like physiological and behavioral responses and is mainly expressed in the paraventricular hypothalamic nucleus (PVN). Urocortin 1 (Ucn1) is also a ligand to CRF type 1 and 2 receptors that has been associated with the stress response. Ucn1 neurons are primarily found in the Edinger–Westphal (EW) nucleus. It has been previously proposed that CRF and Ucn1 differently modulate stress responses to distinct types of stressors. The present study used male Wistar rats to compare the effects of acute restraint stress and unpredictable chronic stress (UCS) through Fos-immunoreactivity (Fos-ir) on CRF-containing neurons of PVN and Ucn1-containing EW centrally projecting neurons. Results showed that PVN neurons responded to both acute restraint and UCS. Also for the PVN, unspecific variables, dependent on the time animals remained in the laboratory, do not seem to alter Fos-ir, since no significant differences between acute and chronic control groups were found. On the other hand, EW neurons were only activated in response to acute restraint stress. Also, for this nucleus a significant difference was found between acute and chronic control groups, suggesting that unspecific variables, dependent on the time animals remain in the laboratory, interfere with the nucleus activation. These results suggest that CRF/Ucn1 neuronal circuits encompass two interconnected systems, which are coordinated to respond to acute stressors, but are differentially activated during chronic unpredictable stress.

Haemodynamic effects, safety, and pharmacokinetics of human stresscopin in heart failure with reduced ejection fraction†

Human stresscopin is a corticotropin-releasing factor (CRF) type 2 receptor (CRFR2) selective agonist and a member of the CRF peptide family. Stimulation of CRFR2 improves cardiac output and left ventricular ejection fraction (LVEF) in patients with stable heart failure (HF) with reduced LVEF. We examined the safety, pharmacokinetics, and effects on haemodynamics and serum biomarkers of intravenous human stresscopin acetate (JNJ-39588146) in patients with stable HF with LVEF ≤ 35% and cardiac index (CI) ≤ 2.5 L/min/m(2). Sixty-two patients with HF and LVEF ≤ 35% were instrumented with a pulmonary artery catheter and randomly assigned (ratio 3:1) to receive an intravenous infusion of JNJ-39588146 or placebo. The main study was an ascending dose study of three doses (5, 15, and 30 ng/kg/min) of study drug or placebo administered in sequential 1 h intervals (3 h total). Statistically significant increases in CI and reduction in systemic vascular resistance (SVR) were observed with both the 15 ng/kg/min (2 h time point) and 30 ng/kg/min (3 h time point) doses of JNJ-39588146 without significant changes in heart rate (HR) or systolic blood pressure (SBP). No statistically significant reductions in pulmonary capillary wedge pressure (PCWP) were seen with any dose tested in the primary analysis, although a trend towards reduction was seen. In HF patients with reduced LVEF and CI, ascending doses of JNJ-39588146 were associated with progressive increases in CI and reductions in SVR without significant effects on PCWP, HR, or SBP. Urocortins Improve Dystrophic Skeletal Muscle Structure and Function through Both PKA- and Epac-Dependent Pathways

In Duchenne muscular dystrophy, the absence of dystrophin causes progressive muscle wasting and premature death. Excessive calcium influx is thought to initiate the pathogenic cascade, resulting in muscle cell death. Urocortins (Ucns) have protected muscle in several experimental paradigms. Herein, we demonstrate that daily s.c. injections of either Ucn 1 or Ucn 2 to 3-week-old dystrophic mdx(5Cv) mice for 2 weeks increased skeletal muscle mass and normalized plasma creatine kinase activity. Histological examination showed that Ucns remarkably reduced necrosis in the diaphragm and slow- and fast-twitch muscles. Ucns improved muscle resistance to mechanical stress provoked by repetitive tetanizations. Ucn 2 treatment resulted in faster kinetics of contraction and relaxation and a rightward shift of the force-frequency curve, suggesting improved calcium homeostasis. Ucn 2 decreased calcium influx into freshly isolated dystrophic muscles. Pharmacological manipulation demonstrated that the mechanism involved the corticotropin-releasing factor type 2 receptor, cAMP elevation, and activation of both protein kinase A and the cAMP-binding protein Epac. Moreover, both STIM1, the calcium sensor that initiates the assembly of store-operated channels, and the calcium-independent phospholipase A(2) that activates these channels were reduced in dystrophic muscle by Ucn 2. Altogether, our results demonstrate the high potency of Ucns for improving dystrophic muscle structure and function, suggesting that these peptides may be considered for treatment of Duchenne muscular dystrophy.

ACTH response to desmopressin in a patient with acromegaly; Expression of corticotropin-releasing factor, urocortins and vasopressin V1b receptor in GH-producing pituitary adenoma

GH-producing pituitary adenomas frequently co-produce other certain anterior pituitary hormones, such as prolactin (PRL). In contrast, GH-producing adenomas which express all of corticotropin-releasing factor (CRF), urocorin1 (Ucn1) and urocortin3 (Ucn3) have not been reported. A 39-year-old woman was admitted for evaluation of the pituitary tumor. The diagnosis of acromegaly was confirmed by elevated serum GH and IGF-I levels, and the absence of GH suppression by oral glucose tolerance test. ACTH response to desmopressin (DDAVP) was observed (plasma ACTH levels increased from 13.9 to 50.4 pg/ml at 90 min). Although it is known that ACTH response to DDAVP is considerably useful for the diagnosis of ACTH-dependent Cushing's syndrome, the diagnosis of Cushing's disease was not supported by the criteria. The patient underwent transsphenoidal resection of the pituitary tumor. Immunohistological examination confirmed a GH- and PRL-producing adenoma, whereas ACTH was negative. ACTH response to DDAVP disappeared after tumor removal. To determine the cause of preoperative ACTH response to DDAVP, we examined expression of CRF family peptides and vasopressin V1b receptor in the pituitary adenoma by immunohistochemistry. Immunohistochemistry revealed positive immunostaining for CRF, Ucn1, Ucn3 and vasopressin V1b receptor in the adenoma. These observations raised the possibility that DDAVP caused an ACTH response, perhaps via the paracrine effects of tumor-derived CRF and Ucn1. When ACTH response to DDAVP is observed in patients with pituitary tumor, not only the direct effect of DDAVP on ACTH secretion, but also a possible involvement of CRF and/or urocortins expressed in the pituitary adenoma, should be considered.

Differential regulation of gonadotropin-releasing hormone by corticotropin-releasing factor family peptides in hypothalamic N39 cells

Corticotropin-releasing factor (CRF) is involved in a variety of physiological functions including regulation of hypothalamo–pituitary–adrenal axis activity during stressful periods. Urocortins (Ucns) are known to be members of the CRF family peptides. CRF has a high affinity for CRF receptor type 1 (CRF1 receptor). Both Ucn2 and Ucn3 have very high affinity for CRF receptor type 2 (CRF2 receptor) with little or no binding affinity for the CRF1 receptor. Gonadotropin-releasing hormone (GnRH) is known to be involved in the regulation of the stress response. Gonadotropin-inhibitory hormone (GnIH) neurons interact directly with GnRH neurons, and the action of GnIH is mediated by a novel G-protein coupled receptor, Gpr147. This study aimed to explore the possible function of CRF family peptides and the regulation of GnRH mRNA in hypothalamic GnRH cells. Both mRNA and protein expression of the CRF1 receptor and CRF2 receptor were found in hypothalamic GnRH N39 cells. CRF suppressed GnRH mRNA levels via the CRF1 receptor, while Ucn2 increased the levels via the CRF2 receptor. Both CRF and Ucn2 increased Gpr147 mRNA levels. The results indicate that CRF and Ucn2 can modulate GnRH mRNA levels via each specific CRF receptor subtype. Finally, CRF suppressed GnRH protein levels, while Ucn2 increased the levels. Differential regulation of GnRH by CRF family peptides may contribute to the stress response and homeostasis in GnRH cells.

The role of CRF family peptides in the regulation of food intake and anxiety-like behavior.

Corticotropin-releasing factor (CRF) and the urocortins (UCN1, UCN2, and UCN3) belong to the CRF family of peptides and are the major regulators of the adaptive response to internal and external stresses. The actions of CRF and UCNs are mediated through two receptor subtypes: CRF receptor 1 (CRFR1) and CRFR2. Their physiological roles, among other functions, include the regulation of food intake and anxiety-like behavior. In this review, we describe the progress that has been made towards understanding how anxiety- and depression-like behavior and food intake are regulated by CRF, UCN1, UCN2, and UCN3.

Corticotropin-releasing factor family and its receptors: pro-inflammatory or anti-inflammatory targets in the periphery?

The corticotropin-releasing factor (CRF) family of neuropeptides is composed of CRF, urocortin 1 (UCN 1), UCN 2 and UCN 3, which can bind to CRF-binding protein (CRF-BP) and two known receptors, CRFR1 and CRFR2, to perform many pathophysiological functions, including inflammation. In contrast to its anti-inflammatory effect in the central nervous system, the roles of the CRF family and its receptors in the periphery are diverse. However, the biological activities of the CRF family in inflammation are circumstantial and remain controversial. It is suggested that this diverse effects of the CRF family may be due to the different types of CRF receptors, different cells and tissues and even different concentrations of CRF family peptides. Though there are difficulties or limitations in establishing the exact modulatory roles of the CRF family and its receptors in peripheral inflammation, the application of CRF receptor agonists or antagonists to treat inflammatory diseases is being pursued with increasing interest. This review describes the effects and mechanisms of the CRF family and its receptors in inflammation; the possible application of CRF receptor agonists or antagonists in inflammatory diseases or disorders will also be discussed.

Identification of members of the gonadotropin-releasing hormone (GnRH), corticotropin-releasing factor (CRF) families in the genome of the holocephalan, Callorhinchus milii (elephant shark)

The gonadotropin-releasing hormone (GnRH) and corticotropin-releasing family (CRF) are two neuropeptides families that are strongly conserved throughout evolution. Recently, the genome of the holocephalan, Callorhinchus milii (elephant shark) has been sequenced. The phylogenetic position of C. milii, along with the relatively slow evolution of the cartilaginous fish suggests that neuropeptides in this species may resemble the earliest gnathostome forms. The genome of the elephant shark was screened, in silico, using the various conserved motifs of both the vertebrate CRF paralogs and the insect diuretic hormone sequences to identify the structure of the C. milii CRF/DH-like peptides. A similar approach was taken to identify the GnRH peptides using conserved motifs in both vertebrate and invertebrate forms. Two CRF peptides, a urotensin-1 peptide and a urocortin 3 peptide were found in the genome. There was only about 50% sequence identity between the two CRF peptides suggesting an early divergence. In addition, the urocortin 2 peptide seems to have been lost and was identified as a pseudogene in C. milii. In contrast to the number of CRF family peptides, only a GnRH-II preprohormone with the conserved mature decapeptide was found. This confirms early studies about the identity of GnRH in the Holocephali, and suggests that the Holocephali and Elasmobranchii differ with respect to GnRH structure and function.

Together with the recently discovered members of the CRF peptide family, urocortin 1, urocortin 2 and urocortin 3, it also has neurotransmitter actions.Previous publication has demonstrated that stress induces CRF release in the paraventricular nucleus of the hypothalamus and the release of both CRF and GABA in the amygdala. Accordingly, the aim of the present study was to determine the effects of the members of the CRF peptide family on GABA release from the amygdala by using an in vitro superfusion system. In order to study the participation of different CRF receptors (CRF1 and CRF2) in this process, rat amygdalar slices were pretreated with selective CRF1 and CRF2 antagonists.CRF and urocortin 1 significantly increased the release of [3H]GABA from the slices following electrical stimulation, whereas urocortin 2 and urocortin 3 were ineffective. The actions of CRF and urocortin 1 were blocked by the selective CRF1 receptor antagonist antalarmin, but were not inhibited by the selective CRF2 receptor antagonist astressin 2B, both administered in equimolar doses. Our results demonstrate that the release of GABA from the amygdala is mediated by CRF and urocortin 1 through the activation of CRF1 receptors.
